Breakthrough technology for beyond simple automation
Representing a departure from conventional, labor-intensive and time-consuming sample preparation methods, the disruptive Xceler8 Technology allows users to extract and purify nucleic acids from a variety of human, bacterial and viral samples at the push of a button.
Overview
Unlike typical commercial spin-column or magnetic-bead technologies, the automated Xceler8 Technology uses a novel four-step chemistry-based approach to purify nucleic acids.
Cell lysis
Once the sample has been added and the X8 Cartridge inserted into the Instrument, the sample is mixed with a lysis buffer and loaded into the channel. Incubation with the buffer lyses the cells to liberate the nucleic acids.
Selective binding
A reversible cross-linker then selectively binds and gently clusters the nucleic acids on to the solid surface of the main channel, forming a loose 3-D network.
Wash away impurities
Impurities and cellular debris are then washed away and sealed in a waste reservoir within the cartridge, leaving behind purified nucleic acids, thereby simplifying clean-up and minimizing contamination.
Elute nucleic acid
The purified nucleic acids are then released from the surface of the channel and eluted into the collection tube, now ready for a wide range of research and clinical applications.
Automated HMW DNA extraction with the X8 HMW DNA Cartridge Kit
HMW DNA extraction performance using X8. Pulse-Field Gel Electrophoresis (PFGE) analysis of HMW DNA extracted from three different sample types using the automated X8 HMW DNA Cartridge Kit. Lanes1-5, replicates of 2×106 PBMC. Lanes 6-10, replicates of frozen Rat Liver tissue (20 mg). Lanes 11-13, replicates of 1×106 HeLa cells (UHMW protocol) and Lanes 14-16, replicates of 1×106 HeLa cells (HMW protocol). M= CHEF DNA Size Marker, 0.2-2.2 Mb.
High RNA Yield with the X8 Cellular RNA Cartridge Kit
Cellular RNA extraction performance using X8. RNA was isolated in parallel from 6 samples of HeLa cell pellets containing 1 X 106 HeLa cells each. The quality of the total RNA was determined with the Agilent 2100 Bioanalyzer and Nanodrop. Elution volume was 80-90 μl.
High DNA yield with the X8 Genomic DNA Cartridge Kit
Genomic DNA extraction performance using X8 Vs. competitor manual kits. Agarose gel analysis of purified DNA from cell culture, gram- & gram+ bacterial cultures; Quantified DNA yield & RT-PCR data from various gram+ bacterial cultures; Ct values (inversely proportional to yield).
Tissue DNA extraction performance using X8. Analysis of DNA isolated from frozen Rat Tail, Liver, Heart and Lung tissue respectively, using a manual kit from Company Q and replicates of automated X8 Tissue DNA Cartridge Kit.
